The news was announced by Nick Grimshaw on the station's Breakfast Show.
"It's really exciting. I love festivals and I love playing them," said Florence Welch, front-woman of Florence and the Machine.
The appearance will be the band's third performance at the Big Weekend having previously played at events in Hackney and Bangor. In Norwich, they will play the main stage on Saturday.

"It's really nice to play outdoors and see the sky.I like performing when it's raining."
This year's event will take place in Norwich's Earlham Park on May 23-24. Full line-up details will be announced on Radio 1 in the coming months, though the station is understandably focusing its marketing on American superstar Swift, who will play on Sunday.
"They are rowdy, insane, in the best way - there seems to be a lot of jumping, which I love," the singer said after being announced in January.
"Everyone talks about the festivals in England being the best ones you can go to. I feel like I've been missing out - I'm really excited."
The Big Weekend has a history of attracting the biggest pop acts on the planet. Last year's event in Glasgow was opened by One Direction and headlined by Coldplay and Katy Perry.
In the lead up to the event, Radio 1 and BBC Learning will open Radio 1's Academy, hoping to aspire to local 16-19 year olds to attend radio shows, master classes and Q&A's.
